Abstract To further understand the mode of action virginiamycin (VM), an in vitro rumen fermentation study was conducted to determine effects VM on and methane production using lactation diets. is approved for control liver abscesses United States preventing or treating ruminal acidosis other countries. Three ruminally cannulated Jersey heifers were adapted 53:47 forage concentrate ratio diet two weeks before use as fluid donors. The treatments (CON) (25 ppm). Diet dried, ground 1mm used substrate. Rumen collected hours after feeding. Substrate (0.5 g) inoculated with 100 mL a 3:1 McDougall’s buffer:ruminal mixture 24 h at 39°C constant agitation. Each treatment run triplicate three runs. Data analyzed mixed model including fixed effect experimental random SAS software 9.4. Statistical significance set when P≤0.05. Maximum gas less (P < 0.01) compared CON (100 vs. 130 mL/g incubated DM). tended reduce per unit digested DM (6.71 9.88 mg/g DM; P=0.07) (4.40 7.16 P=0.06). increased propionate molar % (21.4 16.2%, P< 0.01). reduced acetate:propionate (2.84 3.85, P = DMD (65.5 72.5%, P=0.02) without reducing total VFA concentration CON. increase pH (6.73 6.68, P=0.07). production, proportion, maintained system early diet.
